Understanding Tobacco’s Battle with Insect Pests
Tobacco (Nicotiana tabacum) isn’t just the world’s favorite controversial leaf—it’s a plant scientist’s best friend thanks to its use as a model organism. But even this all-star is no match for some hungry bugs like aphids (Myzus persicae) and cotton bollworms (Helicoverpa armigera). These pests have a knack for making a salad out of tobacco leaves, and until now, the molecular drama behind the plant’s response has been a bit of a mystery.
Researchers just took a deep dive into this leafy soap opera. Using RNA-seq on tobacco plants before and after insect exposure (at 0, 6, and 24 hours), they found that cotton bollworms triggered more genetic changes than aphids. The most exciting action happened in genes related to hormone signaling, secondary metabolism, and transcription factors. JIH1: The Tiny Gene With a Big Job
Jasmonoyl-L-isoleucine hydrolase 1 (JIH1) came out as a superstar gene, ramping up its activity after insect attacks. Scientists used CRISPR/Cas9 to knock out JIH1 and saw cotton bollworms eating these mutant plants shrank by a whopping 25.67% compared to those munching on regular tobacco. The mutant plants also juggled their growth and defense hormones differently—activating jasmonic acid (JA), salicylic acid (SA), and ethylene (ET) pathways, growing shorter but with longer roots. Most of the JAZ genes (13 out of 15) got silenced in these plants, showing just how central JIH1 is in the tug-of-war between growing tall and fighting off pests.
Of course, not every JIH1 mutant was a hero—some plants didn’t get tougher, probably because of evolutionary quirks in tobacco’s genetics. But overall, this research reveals that JIH1 is like the plant’s personal gym coach, helping it decide when to grow and when to flex its defense muscles.
